Jam tomorrow, jam yesterday, but never ever jam today…

8 December 2006 by kristarella

I hope today is a bit better than yesterday. I don’t know where my head was at yesterday but things were just going wrong. I put the wrong amount of water in one of my PCRs, it was only about 0.5 microlitres (per reaction) more, but then if you add that to when I put the mix into each reaction and put 0.5 microlitres less into each one, you can see my brain is not functioning optimally.

Later I was doing some RNA extraction from kidney tissue and I put the chloroform in (which is supposed to keep the RNA in an aqueous, i.e. watery or dissolved, phase) and I added isopropynol at the same time, which is supposed to precipitate the RNA (make it solid again), so the two opposite thing. Anyway, I think it was mostly fixed except for one sample.

Even later, I was putting the names in a container for Kris Kringle at work and when all but three people had drawn a name there were only two bits of paper left. I’d definitely written all the names, after we had done it all again I found the other bit of paper on the floor . 🙁

I ended up working until 6 (very uncharacteristic of me) and just missed the 6:10 bus. The 6:20 bus never came and the 6:30 was 5 or 10 minutes late. I just missed the 6:45 train so I had to catch an all stops, not as comfortable but thankfully didn’t take to long. It was getting to about 8pm so I stopped to get pizza, that took about 25 minutes… when I got home it was pretty sweet, sat down in front of the TV with my pizza and beer.

Gotta run, meeting someone for coffee!

Had a great Saturday at the GKR Karate State Title Had a great Saturday at the GKR Karate State Titles! I was a sub for @jamesxuereb.me in the Blue Flame Dragons thanks to his sprained ankle; we won first round and came fourth overall!
I did a bunch of judging and officiating, which was really good.
I didn’t place in my individual events, but had a very fun final round of kumite (sparring).
